Abstract

H-Phosphonate synthesis of oligodeoxynucleotids is described in which acetonitrile is replaced by dichloromethane. This solvent is thereafter easily regenerated and reused. Practically "one solvent" procedure of oligonucleotide synthesis has been established. The preparation of 5'-<i>O</i>-(4,4'-dimethoxytriphenylmethyl)-<i>N</i><sup>2</sup>-(<i>N,N</i>-dimethylaminomethylene)-2'-deoxyguanosine (<i>Ic</i>) and 5'-<i>O</i>-(4,4'-dimethoxytriphenylmethyl)-<i>N</i><sup>6</sup>-(<i>N,N</i>-dimethylaminomethylene)-2'-deoxyadenosine (<i>Id</i>) is described.
